Looking forward to discuss some of yesterday’s provocative statements by Peter Verhaert at his flashtalk (below) at poster #67 at the European School of Molecular Medicine (SEMM) workshop in Milano later today:
- formaldehyde fixation is good sample prep for biologically relevant MS imaging
- paraffin embedding is not bad either
- historic, archived, pathologically well documented FFPE samples from tissue banks all over the globe can be analyzed by MSHC (mass spec histochemistry)
- this could render some preclinical disease models redundant
- atmospheric pressure ionization (MALDI as well as DESI) high-resolution MSHC work on histological sections on regular microscope glass slides
- MSHC is spatial, multiomic, top-down, label-free, enzyme-free, antibody-free, and, therefore, elegant, simple and cost-effective
- MSHC does not include extensive (antigen retrieval) washes, therefore not delocalizing small molecules
- MSHC localizes endogenous peptides & (lipophobic) metabolites
- MSHC does not yield extremely high numbers of biomarker candidates, but these numbers are not always necessary?
- MSHC is well-aligned with today’s pathology practice.
