Process development always involves trade-offs. Push for higher yield and your cost creeps up. Optimize for cost and you risk falling short on the cells your therapy actually needs. So which process condition is "the best"? It depends on what you're willing to trade. This is where multi-objective optimization earns its keep. Instead of one answer, you get a Pareto front, the set of process conditions where you cannot improve one objective without giving up on another. Set a minimum yield, see what it costs you. Tighten your cost ceiling, see what yield is still on the table. The decision stays with you, but now it's an informed one. Generating this Pareto front requires screening thousands of process conditions, which is not feasible experimentally. Instead, we use a custom hybrid model of the bioprocess, coupled to a cost-of-goods model, to perform in silico experiments and explore all conditions within minutes. For our hTERT MSC project, that meant 5,000 simulated conditions, collapsed down to a handful of Pareto-optimal solutions.
