Elacestrant in Combination with Everolimus for Estrogen Receptor-Positive, HER2-Negative Previously Treated Advanced Breast Cancer: Results from ELEVATE
A phase 1b/2 umbrella trial evaluated elacestrant plus everolimus in ER+/HER2− advanced #BreastCancer after progression on endocrine therapy 💊 and CDK4/6 inhibition. Among 50 phase 2 patients, median PFS was 8.3 months, with clinically meaningful activity regardless of ESR1 or PIK3CA mutation 🧬 status. Most adverse events were grade 1–2, with low rates of treatment withdrawal or dose reduction.
✅ The findings support elacestrant as a potential oral endocrine backbone for pathway-directed combinations.
